How Much Cake Mix To Use For Round Cake Pans

Cut on sliding microtome

The advantage of a sledge microtome is the flexibility one has with the knife guide. The rotary microtomes
can be varied usually the inclination (the tendency to block surface) of the knife, but when the declination we seldom have much leeway.

The sliding microtome can cause extreme declination of the knife almost parallel to the object, which is a very slow penetration of the knife blade to prevent due to deformation or tearing of the section.

sharpen microtome:

important for successful cutting is first a sharp microtome. The
Approach to sharpening a knife is already at Romeis, Microscopic Technology, 1968, page 123, § 428 admirably described
must "To pull off and grinding of the microtome knife can be slid onto his back, an extractor, ie a tube-like Blind metal sleeve, the length should be equal to the length of the knife,.. d-diameter require a special puller It is best to use a 36cm long strop with upholstered pad, set at a at the table screw-clamp placed the initial third of the belt with red, grobgekörnter grinding paste , the middle rubbed with black fine-grained paste, the last Third remains untreated. The knife, which is provided with puller and grip, then back forward, first on the red surface of the belt under slight pressure, without printing out later, turned over the back and then returned to the starting position. This so-called frontloading is complete when the knife blade under microscopic control completely straight and has more than a very fine perforation. Then the knife is drawn to the leather section, but now always without pressure and not too long. For a complete deducted knife edge is perfectly straight and without perforation, is the last cut by 2-4 times pulled off (not more often! polished) carefully on an untreated part of the leather belt. It's best course of three separate strop used. is most frequently absent due to long pulling off, so that the edge is rounded. From time to time have the Mikrotomesser be ground "

I use three separate strop.

cutting on the microtome:

Sliced, a piece of branch of Abies nordmanniana
The. object was first fixed in a few weeks AFE, the acetic acid content was increased.

The blend is classic AFE from: ig 5ml 40% formol, 5 ml acetic acid and 90 ml 70% ethanol.
For the Nordmann fir, I modified the mix: 40% strength formalin 10ml, 10ml and 80ml acetic acid 70% ethanol.

softened just fixing mixture of acetic acid content in the woody parts and makes the object be cut. Such previous attempts to Tilia with only 50% ethanol fixed root pieces showed me the clear improvement of the cuttability by acetic acid in the AFE mixtures.

After a sufficiently long residence time in the AFE (several weeks) I have the fixing mixture by placing the objects in tap water - changed several times over a few days away - Wash and store in Strasbourg on out mixture. This should still be changed several times in order to get any residual acid that may damage the blade when cutting away.
from the Strasbourg mixture can be gathered from the objects and cut directly into the clamp and sliding microtome:

on the knife a ~ 70 'ethanol solution is applied, so that the section can float and not to tear through sticking. If
can roll up the cuts with a brush to prevent this by hangs up without pressure the hair of the brush on the stem and then with the knife through the cut out.

has the ideal angle of inclination to try out - I always think the extractor to the knife when the knife is clamped in the microtome, and draw a line from the Mikrotomspitze to the lower, outer edge of the imaginary Puller - this line should be a few different angles from the horizontal line up.

The result:

The sections were first bleached in sodium hypochlorite and repeatedly washed in tap water. Then, the sections for about 15 minutes Etzold colored in green and isopropyl alcohol and xylene and embedded in Malinol:

Funny Writing On Birthday Cake

Start the Glassfish Admin Console accelerate

[UPDATE]
There is also an official way to speed up the Start:

http://www.mentby.com/Group/glassfish-users/glassfish-v3-admin-console-very-slow.html

This is monitored when the Glassfish in domain.xml option

\u0026lt;jvm-options>
-Dcom.sun.enterprise.tools.admingui.NO_NETWORK = true
\u0026lt;/ to set jvm-options>

and the file

GlassFish / modules / console-update-center plugin.jar

whatsoever. Then still in the domain of the two directories
  • osgi-cache
  • generated

delete and start the Glassfish again.
[/ UPDATE]

When you call the admin console of Glassfish, it sometimes takes a long time until it opens. The reason for this are not extensive initialization - the CPU load is at 0 while - but a "phone home" to check for updates. You can avoid this by inserting in / etc / hosts this entry:

127.0.0.2 pkg.sun.com pkg.glassfish.org

So the admin console launches much faster.

Lifespan People With Wagners

portrait photography - portraits of women - beauty retouching

In this paper I discuss skin and beauty retouching in portrait photography. The skin you can work with many different Photoshop tools. I always edit the skin in 2 steps.
In step 1, remove I have the rough skin of impurities and in step 2 is the fine tuning that makes the skin appear softer. Below I will discuss the tools usually used by me and describe their use.

Before I begin with the retouching, I first create a copy of the current image. This way I can quickly switch between the original and the edited version change. The copy I create "Create New Layer on the bottom right. One can this simply drag the image to the icon. Create
New Layer
first Step 1 - Clean the skin
In this step, I am dealing with the removal of skin blemishes, spots and shiny skin. Moles and scars I usually retouch away not because they are distinguishing characteristics of the person and make their own charm can. Ultimately, however, it is a matter of taste.

The actual tools I use are in Photoshop on the left.

1.1 Patch tool
The Patch tool can determine a user-defined selection area and copy it to another location. The tool takes into account the original pixels and attempts to integrate seamlessly copied source pixels. Generally you should select source pixels, the original pixel color are similar and present the same texture. Thus, the retouched area falls to less later.
Patch tool
I use the Patch tool like to remove blemishes. On the lower screen shots of the individual steps are shown.
Patch tool - Step 1
Patch tool - Step 2

Patch tool - Step 4
2.1 Healing Brush tool
Another handy tool for removing skin blemishes, the Healing Brush. It works much like the Clone Stamp tool, which I will explain later. Unlike the Clone Stamp texture, lighting, shading, and alignment of the korrigiernden areas and source pixels to be considered.
Healing Brush tool
with the right mouse button you can open the context menu of the brush and settings. Most relevant here is the hardness of the brush. For surfaces with no clear edges (like skin), the brush should be set quite soft. In this way one obtains a smooth transition up between the original / retouched areas.
Healing Brush tool - Settings
determined by holding down the Alt key is the output range (puck), of which included the source pixel should be. Then you lässst release the Alt key and starts to paint over the blemishes by clicking on the appropriate location.

1.3 Clone Stamp tool
The Clone Stamp can copy source pixel 1:1 and move to another location. In addition, the clone stamp various modes by which you can change the display of the copied pixels. In this example, I use the clone stamp to correct shiny skin.
Clone Stamp tool
submode choose Darken and Opacity the transparency of the die determines about 20%. Using the right mouse button to open it, like the Healing Brush from the context menu, for example, to determine the hardness.
Clone Stamp tool - Settings
By holding down the Alt key take on source pixels from a darker place the forehead. Then you lässst release the Alt key and starts the "stamp of" the shining face section by moving the mouse over the appropriate places. This process is repeated until the desired result was achieved up.
Clone Stamp tool - Darken
The best is to experiment with the above mentioned tools around to achieve the desired results. The figure below shows the comparison between the original and the retouched version. There are blemishes, pimples and removes shiny patches.
vs original. Beauty Retouching - Step 1
second Step 2 - make skin soft
First I create again a copy of the Create New Layer. Then I go to Filter in the top menu and choose among "Blur filter" the "Gaussian Blur ..." from.
Gaussian Blur
About radius determines how much you want the image to be blurred. In this example I choose a higher value.
Gaussian Blur - Settings
Then I create a layer mask on "Add layer mask".
Add Layer Mask
About the "Paint Bucket" on the left side under Tools, you do your white layer mask black to make the original retouched visible. Then you select the Tools normal "brush tool" with the color white. If you paint on the black layer mask with white, the blurred image to be visible. In this way one can represent a targeted only the skin soft and highly detailed features such as hair, eyes can focus. The last step is determined by the opacity of the levels, how much effect the blur effect.
The following screenshots document once all the above steps.
Blur - Step 1
Blur - Step 2
Blur - Step 3
Blur - Step 4
The result and the comparison between the original and the version with beauty retouching you see in the image below.

Finally, I've listed a few websites that also deal with the issue of skin retouching :
